Here's the updated question: Can a Holosun 507C optic be mounted DIRECTLY ON THE FACTORY OPTIC READY SLIDE OF THE P320 M18 WITHOUT ADDITIONAL MILLING? I am pretty sure that you will need to use the aftermarket adapter plate

Peter, you will not have to use an adapter plate.  RMR for which the slide is milled for and the Holosun are the exact same dimensions down to the millimeter.

Peter, You will have to use an adapter plate to mount the 507C to the optics milled slide of the M18. Springer precision, OuterImpact are options.
