Yes to 45 Long Colt rounds to be specific. You can watch YouTube videos of this exact gun firing 45lc rounds.
Also, if you shoot a bunch of the 45 colt in a 454 chamber pay attention to cleaning the chamber before you shoot the 454, a ring of carbon and other material can build up in front of the case mouth. Clean the chamber real well and you are golden.
