Hi James and welcome. Standard Saami Pressure .45LC ammunition (new) will work no issue as well as Cowboy loads. Buffalo Bore/Grizzly Hunting loads should be avoided for safety and warranty reasons. Cheers and stay safe!

General concensus on the Ruger forum is that the Vaquero can't handle the Grizzly loads.
