The cartridges load one at a time through the loading gate so moon clips are not feasible. You can use 45 ACP which head space on the case mouth or 45 Auto Rim which headspace on the rim.
no half moon clips required. cylinder is machined so that the cartridge headspaces on lip (in cylinder) just like in the barrel of a semiauto I have the Blackhawk 5.5 in version and they are great

No moon clips needed. The 45ACP head spaces on the cartridge case (the exposed part around the bullet). This pistol also utilizes Ruger's New Vaquero reverse indexing pawl ( On most Ruger single actions, as you load and unload, it is possible to turn the cylinder a fraction too far, leaving the charging hole out of alignment with the loading gate, and you can't turn it back. You have to keep turning the cylinder, loading or unloading the remaining chambers until the one you skipped comes back up again. With reverse indexing, you can rotate the cylinder back and not have to skip that chamber.
